Vildagliptin is a DPP-IV inhibitor for antihyperglycemic research

**Background**

Diabetes mellitus is a chronic metabolic disorder characterized by hyperglycemia resulting from defects in insulin secretion, insulin action, or both. A key component in the regulation of glucose homeostasis is the incretin system, which involves hormones such as glucagon-like peptide-1 (GLP-1) and glucose-dependent insulinotropic polypeptide (GIP). These incretins stimulate insulin secretion from pancreatic beta cells in a glucose-dependent manner. However, their biological activity is rapidly terminated by the enzyme dipeptidyl peptidase IV (DPP-IV), which cleaves the active forms of these hormones. Consequently, the inhibition of DPP-IV has emerged as a significant therapeutic strategy to prolong the half-life of active GLP-1, thereby enhancing insulin secretion and improving glycemic control. In this context, we will introduce a potent and selective DPP-IV inhibitor – Vildagliptin.

**Definition**

Vildagliptin is a potent, stable, and selective dipeptidyl peptidase IV (DPP-IV) inhibitor with an IC50 value of 3.5 nM in human Caco-2 cells.

**In Vitro and In Vivo Studies**

According to the Vildagliptin description, this compound possesses excellent oral bioavailability and potent antihyperglycemic activity. Vildagliptin in vitro studies have demonstrated that the compound promotes the survival of pancreatic beta cells by inhibiting cell apoptosis and promoting cell proliferation. Regarding Vildagliptin In Vivo activity, research using male db/db mice (BKS) and wildtype mice showed that administration of Vildagliptin (35 mg/kg; once daily by oral gavage for 6 weeks) significantly increased plasma active GLP-1 levels (22.63±1.19 vs. 11.69±0.44). Furthermore, in obese male Zucker rats, oral administration of Vildagliptin (10 μmol/kg) significantly decreased glucose excursions and stimulated insulin secretion. These results highlight the efficacy of the compound in managing glucose levels in diabetic animal models. For researchers requiring specific Vildagliptin technical information, these findings underscore its role as a critical tool for studying incretin-based therapies. In conclusion, Vildagliptin is a potent DPP-IV inhibitor that effectively enhances GLP-1 levels and protects beta cell function.
